As previously discussed, when leaky gut occurs, the intestinal lining becomes permeable, allowing pathogens to escape into the bloodstream, triggering an immune response and widespread inflammation. This gut-derived inflammation can compromise the integrity of the blood-brain barrier (BBB), leading to what is known as leaky brain.
Toxins, harmful bacteria, and other pathogens can then infiltrate and influence the brain, potentially triggering the onset of various diseases in individuals with genetic predispositions, including depression, Alzheimer’s, Parkinson’s disease, and other memory and mood disorders.
Once inflammation sets in within the brain, neural messages may not be transmitted as efficiently, resulting in brain fog and symptoms such as poor memory, fatigue, depression, and anxiety.
The Role of Microglia
Microglia, the immune cells of the brain, diligently maintain the microenvironment of the central nervous system (CNS) by eliminating potential toxic molecules. Comprising 10-15% of all brain cells and outnumbering neurons by a ratio of 3:1, these resident macrophage cells serve as the primary immune defence within the CNS.
Initially neuroprotective, microglia safeguard the blood-brain barrier from damage caused by systemic inflammation. However, when infectious agents and pathogens breach the BBB, inflammatory cytokines are activated, prompting microglial cells to swiftly react to reduce inflammation and eliminate the invaders to prevent harm to delicate neural tissue.
Activation of Microglia
Under normal circumstances, microglia function to protect neurons. Yet, disruption to the BBB and subsequent heightened inflammation triggers a notable shift in their behaviour. Instead of safeguarding neurons, they begin attacking BBB components, compromising its integrity, and damaging neurons.
This cellular damage, resulting from microglial activation and subsequent inflammation, generates neuronal protein fragments. Consequently, the immune system produces antibodies against these damaged cellular proteins, leading to neuro autoimmunity, where the immune system targets its own brain cells. This neurodegenerative process contributes to conditions such as dementia, Alzheimer’s, and Parkinson’s disease.
Your Vagus Nerve Regulates Inflammation via Your Microbiome
Effective communication between the Immune system and the Brain is crucial for managing inflammation, with a key player in this communication being the Vagus nerve. Serving as the primary conduit between the gut and the brain, the Vagus nerve connects many major organs, particularly those within the gut. Its primary role is to relay information from internal organs such as the liver, heart, and lungs to the brain.
The Vagus nerve also plays a vital role in immune system regulation and inflammation control. It transmits anti-inflammatory signals to various parts of the body through three distinct pathways:
- The first pathway involves the HPA axis.
- The second pathway is the splenic anti-inflammatory pathway, where the Vagus nerve stimulates the splenic sympathetic nerve, triggering the release of noradrenaline and acetylcholine.
- The Third pathway is known as the cholinergic anti-inflammatory pathway, which stimulates the release of acetylcholine to inhibit inflammatory molecules.
During periods of chronic stress, the Vagus nerve can activate inflammation and disrupt immune regulation.
In recent years, the Vagus nerve has emerged as a notable target for potential therapeutic interventions, not only in the realms of psychiatric and gastrointestinal disorders but also in addressing various inflammatory conditions like rheumatoid arthritis, chronic headaches, fibromyalgia, and diabetes. It has been observed that disruption of Vagus nerve signalling can exacerbate systemic inflammation in numerous diseases.
The Vagus nerve is profoundly influenced by the presence of bacteria, fungi, and other microbes in our gut microbiome. Certain probiotics, such as Bifidobacterium, have been found to communicate with the brain through vagal pathways. Recent studies have revealed that individuals with depression, anxiety, and schizophrenia exhibit alterations in both their gut microbiota and vagal tone. Your Gut Microbiome – what is the Gut.
The gut is a long tube in the body of a person through which food moves during the process of ingestion, digestion, and assimilation. Your Microbiome is a collection of microbial genomes and microbiota is a collection of microbes It consists of bacteria, viruses, parasites, protozoa, and fungi inhabiting the gut. Estimated one hundred trillion organisms. This family of organism’s weighs +/–2kg (like weight of adult human brain) and ideally optimally functional in a balanced healthy individual. Normal gut flora is essential for the optimal functioning of the gut and for maintenance of general health and wellbeing. “Dis-ease” (a state of dis -ease in your health) only really sets in where “dis-biosis” (a state of dis-biosis in your healthiest of microbiome) sets in affecting and afflicting the entire body.
Functions of the gut microbiome
- Regulates the immune system (responsible for protection and defence against infections)
- Regulates the endocrine system (responsible for production of hormones)
- Seventy percent serotonin produced in the gut by the microbiome (Candida, Streptococcus, Escherichia, Enterococcus) – Serotonin is the “happy hormone” in our body. Decreased serotonin levels in the brain is a major contributing factor for depression.
- GABA, Dopamine, Acetylcholine and Noradrenaline are all made by gut bacteria. These are amino acids and hormones which play essential roles in the signalling pathways of the nervous system.
How do we affect these bacteria?

The gut and brain communicate by molecules carried in the blood, and microbes influence those chemical messages. Microbes also interact with the gut’s special nervous system, called the enteric nervous system. It has a direct, two-way connection with the brain via the central nervous system. The Gut-Bran Axis or link: This is a communication network that links the central nervous system (CNS) and the enteric nervous system (ENS). This axis functions in a bidirectional manner, therefore, each component of the network can influence the other. Hence the gut is known as the second brain. Psychobiotics (prebiotics and probiotics) against stress: a new paradigm. Recent Studies.
The gut microbiota is now identified as a key channel for brain processes, mood, and behaviour. The development of therapeutic approaches as targeted at the gut microbiota will help to improve the mental health of populations. An Irish team studied for 4 weeks, the impact of a psych biotic diet on the gut microbiota, mood, and stress perception. The researchers recruited twenty-four participants and twenty-one healthy adult controls aged 18 to 59 years:
The first group had a diet that included whole grains (5 to 8 servings), fruits and vegetables high in prebiotics (6 to 8 onions, lettuce, cabbage, apple, bananas, oats), other vegetables (3 to 4 servings) and fermented foods (2 to 3 servings of sauerkraut, kefir or kombucha). They were also asked to cut back on treats, sodas, and fast food. Participants also benefited from general dietary advice: knowledge of the food pyramid, daily calorie recommendations according to gender, etc.
Control subjects only received this dietary advice. The stress experienced by all subjects was self-rated by the Cohen Perceived Stress Scale (PSS) at baseline and at the end of the study.
Results after 4 weeks:
– Only the psychobiotic diet group had a significant decrease in perceived stress levels. In addition, the higher the adherence to the plan, the greater the reduction in the PSS score.
– Metabolomic analysis revealed a significant change in forty lipid metabolites in the psych biotic diet group, not in the control group. This could mean that the gut microbiota influences mood by regulating lipid metabolism.
Micro biotic medicine and psych biotic treatments can modulate the gut-brain axis and reduce stress and associated disorders. Dietary interventions must already have their place in nutritional recommendations for the preventive or curative treatment of mental disorders.
